Labcorp Genetics (formerly Invitae), Labcorp RCV001856876 SCV002239700 pathogenic Charcot-Marie-Tooth disease axonal type 2Z 2021-07-20 criteria provided, single submitter clinical testing For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this variant affects MORC2 protein function (PMID: 32693025). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt MORC2 protein function. ClinVar contains an entry for this variant (Variation ID: 423564). This variant has been observed in individual(s) with clinical features of Charcot-Marie-Tooth disease (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with serine at codon 266 of the MORC2 protein (p.Arg266Ser). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and serine.
GeneDx RCV000479173 SCV000573276 likely pathogenic not provided 2019-10-14 criteria provided, single submitter clinical testing Not observed in large population cohorts (Lek et al., 2016); In silico analysis, which includes protein predictors and evolutionary conservation, supports a deleterious effect; Has not been previously published as pathogenic or benign to our knowledge; This variant is associated with the following publications: (PMID: 32693025)
